Done all, just in time too - it gets a little boring at the 6th stage (one can only take so much repetition). Bit disappointed that boss can only be hurt after reflecting a fireball back at him. Hoped bombs would do the same.
It's best to unlock the next area and then move on instead of repeating missions in the earlier areas. Only completing quests in the newest unlocked area will lend to unlocking the next area. Also, the later areas tend to give more gold and give more gold more quickly. This means you can complete the quest Big Spender (buy everything in the shop) faster by reaching the later levels and completing missions there for gold instead of grinding out missions in earlier stages. On top of this the upgrades don't help that much, so the game won't be overly difficult if you head into the later stages without many upgrades.
Lastly, I've noticed that all treasure pieces and assassination targets are always on the last floor of a stage. There is no need to explore earlier floors (unless you are looking for a power-up) when playing those two modes. Simply get to the finally floor and then do what needs to be done.
The quests are equivalent to the difficulty of the game. Very good game for a boredom killing.
As it was mentioned above, it is better to continue unlocking new areas instead of repeating the missions. When you reach the end you will have all the quests eventually.
At some point, the boss starts to shoot fireballs. You need to send them back to him. He becomes vulnerable for a while and that's when you have to attack him. Repeat a few times and he'll die in no time

Don't forget to keep moving aswell. There is big bombs that fall on you and follows you around a bit but it will stop just before it drops. Look for the shadow around you. Hit the red bomb guys away and don't run in to them or try to hit towards boss because they don't damage it. Rest is stated in above post.
