Fairly easy all around. Completed all quests in 2 playthroughs, and that was only because I messed up the first one badly.
Few tips:
1) Do not upgrade ANYTHING until you can afford the Radio Tower. The earlier you can get more survivors, the easier the game will be.
2) For scavenging, more is NOT better. I completed all of the quests with a part of 2 well-rounded scavengers. It seems to function off averages rather than just adding in people. That is, if you have 2 2/2/3 assigned and assign a 1/1/3, your avoid discovery and survival chance will go down significantly. Morale also seems to help this significantly.
3) As soon as you have a spare survivor, always have a survivor assigned as your assistant. He will reload the weapon you aren't firing. I can't even begin to stress how awesome this is. As soon as you get the grenade launcher, the game becomes face-rollingly easy - fire whatever weapon you want, swap to GL, fire a shot, swap back, wait for assistant to reload, swap, fire grenade, swap back.
4) The easiest way to get High Explosives is to wait until the mid-30 days, then buy 3 C4 (NOT mines), lay them out, and trigger them when a big group is over them.
5) If you're having trouble with Marksman, remove survivors you have assigned to shooting. They will kill several zombies over the course of a fight. Even later levels only have about 45-50 zombies, so survivors killing even 5-10 of them makes it much harder.
I think that it can't be completed before wave 35 or so, simply there aren't enough zombies to headshots. Actually, they are enough, but they reach the barricade before that you can headshot them beacuse the fire rate of Remington is too low (unless you're pro and can easily go for a "1 shot-1 headshot" run).
You shouldn't assign survivors to the shooters group, they steal kills and make the things harder. It's better to assign survivors to the melee group so they can defence the barricade from zombies that you miss.
Don't use the Remington's scope, I found easier to aim without it.
Obviously, don't use any other weapons to kill the zombies. If someone reachs the house, you should leave him to melee fighters and focus on other zombies' head.
@bakkart : Try combining all the tips given above? If you use the ones given by @Olinser, @Laspa and @AmemEvol together, you should be able to get it pretty easily... I did it the 'normal' way and it took me 5 tries to get it!

some tips on high explosives.
This can only be done after u buy the explosives lab, and after 30+ days in.
Get the C4, not the mines and place them along the top where the zombies come from. When a huge clump arrives, detonate the explosives and you should have the medal. If you dont get this on your first try, maybe take off any shooters so they dont steal your kill.
so about the furry friend quest, if you don't find him in the 40 days, you have to restart and play again.
You shouldn't have too. There is an unlimited mode after you beat the game that lets you continue searching the city for survivors, supplies, and weapons.
Although, if you search every area in the city and do not find the cat survivor, then you may have to restart the game (but I'm not 100% certain on this).
I found Tough Guy very easy, on my first try I won entire game without getting damaged once. I used mostly single-shot weapons and got as much extra money from headshots as I could, and this weapons are so cool to use. At the beginning I used mostly Bow and Crossbow, after I got more weapons I used Springfield + Remington duo, worked rather nice, at the point I got the grenade launcher it became very easy as Remington + Grenade destroys basically everything.
Marksman was rather difficult, I think it is much better to use Remington only and don't use secondary weapon as it just steals too many kills, better to just reload manually and focus on getting those headshots.
@Reton8 : The unlimited mode is separated from searching the city... once you've searched all the houses and compounds, you can not search anything else, which means that if you haven't found the furry friend during the entire search, you have to start over from scratch. =)
Also... I had a good run during my 5th gameplay and ended up with meeting a total of 25 survivors! (oO)
And to complete it, here are my final stats after my 5th run that awarded me the "The Collection" Quest! =)
