Well if you look at in terms of what they actually do take I would say probably a boss in the Final Fantasy Games. For instance, Sepiroth, not only does he have several stages to his fight, but if you play the game you know you can call a freaking meteor out of the sky to smash into his body which will only take away a fraction of his life. Not mentioning Knights of the Round Table, that move calls upon 13 different Knights to slash away for an entire animated scene, but yet still doesn't take away have of his hit points. So in that sense I say bosses in games like Final Fantasy take the cake in how much they can take
Actually it's 17 colossi. Dang its hard to find a pic of final boss! Probably because he shoots lazers at you and you can't ever get a good look at him while it's so dark!

I gotta go with Nico Bellic from GTA4. He can crash through his windo and fly 100 feet and still get up. not to mention he can take a close up shot gun shot w/o dieing.
i think hancock. i seen the movie and he keeps getting shot but it doesnt even hurt him. 1 part he almiost dies but he still gets back up. he even stopped a train, and blocked a bazooka type thing shot. but he is not in a game so i dont think he counts. if he dont than i think graham is right.
