In order to have this Quest unlock, you need to finish your combo run. Either by letting it run out on its own, or by ending it by clicking the 'End Combo Early' button in the game!
Easiest way to get the "One Upgrade Only" Quest, is to first play a while with getting 80, 100 or 120 per turn hitting the turtles. Make sure you play untill reaching $ 5,000 and then buy the machine gun, never fails, always works!
Allright... I've now officially given up on the 100,000,000 combo Quest!! I played the game for over an hour and reached 2 million combo with turtle flows of 305, 200 and 200, a filled ceiling of lasers, a ground filled with buzz saws and machine guns, the middle filled with bumpers and the biggest mega hammer...
Calculation would then show that, in order to reach the 100M Quest, I would need to let the game run for a least another 50 hours... ... (O.O) ... WUT?!?!?! ... forget it! This one shouldn't be labeled "Hard" but "Impossible"! =(

What kind of computer do you have (processor speed etc.). I did this one in about 12 hours or so on a nearly 3 year old laptop running. [Intel Pentium Duo Core processor 2,20 GHz each, 4 GB Ram, Standard not-so-special graphics card, Windows 7 64-bit. (TMI?)]
Upgrade the turtles to flow 9 on each pipe, this is actually where you get maximum flow. (Upgrade it to 10 and you'll see the flow decrease).
The lasers may also be causing you lag. So in game time it may theoretically faster, but in real time it's just slowing you down.
This is how to get 100 freaking million, fill your screen with buzzsaws groundsaws, lasers, machine guns, stompers, and etc.
I don't know how that set-up will run on a fast computer, but I'm pretty sure stompers don't make the combo go up and faster after a certain point. (I did some calculations).
Also, "One Hundred Freakin Million" showed up on my quest pages. Yay! and thanks!
Honestly, I don't understand this game at all. How am I suppose to get combo's when the hammer swings like every 5 or more seconds. I don't get it. I don't get it. And there isn't any help for the game.
