Hi, I am level 3142, and I am going to be the first to get a grade 100 gem.
How did I get specs obscenely high? Well, thanks to this dude over here...
One important thing to note is how to create dual gems. Adding two grade one's them spamming U is losing about 33% special power.
Optimal creation is a G1 of both, combine, make a G2 of each, add one pure to each dual, combine the G3s, duplicate, and repeat with pure G4s, duplicate the 6, etc.
here is the difference at g34: Regular creation: http://prntscr.com/3dgs2c
Special Mix: http://prntscr.com/3dgs6k
Note that the hit count is the same, and the cost to duplicate is the same. This is a difference of 32.9%, and this difference keeps rising at higher levels. It's only a 16% difference at grade 6. It takes a decent amount of time, but it is worth it at ultimate grades (45+).
I now have huge specs. It takes me about 20-30 seconds real life time to get +1 grade. So a minute before +2(needed if want extra specs). I estimate my specs to be about 80% higher than normal, at least blood is.
PS- Level 46 amps got the spec treatment too, they are wicked.
@oLaudix: I think, your chart is not exact: I have a mixed Gem, started with 32-spec, 16c till G30 or 40, then 64c, and it has 1.3e10 hits and I have 5e30 Mana + the mana spent in Gems, another ~5e30mana...
The Amps are strict 16c.
I don't update the managem anymore, cause the monsters, 1e35 hp - started to take damage...

I dont count for orblets. Even then the ratio for the gems still stands. It is accurate for that. More to that it onlu counts mana leeched. But other variables like mana gained from mobs and earlier waves are equal for all of the situations so it can be omitted. While the specific nr of hits needed is wrong, the ratio of those hits for each situation is same. I wanted to know which GEM gathers mana quicker.
Also mana spent on gems is highly inaccurate. Right now i have gem for 110k in trap +6x3,4k amplifiers and it says is pent 19,539 so you can use it.
@oLaudix can you make a close up of below 1:e10 hits? i would like to know if its an good idea to start with 32-1024 and then continue with 64 or 16 methods. can you check that?
Fine - I think, that could be tranferred to the killgem, too. As I am actually in my 'defreezing' phase, it would be clever to redo my killgem with 32s&64c and amplify it with grade+1 gems... Hmmm - That means, no G100 by now but one G98 with 6 G99... I decide in one or two days, when my cosy monsters are unfrozen
@bilboCGL, your links didn't work because you should use links to image, not to page. You can rightclick on the image and then on "copy image url" or something like that (depends on your browser)
I started with 32s and 64c, at G80 I did 16c resulting in G83 and finaly I pressed just 'u'... I thought about leaving it G99 and enrage with higher grades but I wanted a G100
I just didn't had the patience to freeze some more days - no macro used....
It seems, that my fusion skills are a little higher than yours, when I look at the manacost of your gem. (45+14 IIRC).
btw: Right now I'm coming to the end of a no-mp run, WL 60 and just reached X1 - Interesting, too and it takes less time for me to finish the game than building one G100...
