Hi, I am level 3142, and I am going to be the first to get a grade 100 gem.
How did I get specs obscenely high? Well, thanks to this dude over here...
One important thing to note is how to create dual gems. Adding two grade one's them spamming U is losing about 33% special power.
Optimal creation is a G1 of both, combine, make a G2 of each, add one pure to each dual, combine the G3s, duplicate, and repeat with pure G4s, duplicate the 6, etc.

Note that the hit count is the same, and the cost to duplicate is the same. This is a difference of 32.9%, and this difference keeps rising at higher levels. It's only a 16% difference at grade 6. It takes a decent amount of time, but it is worth it at ultimate grades (45+).
I now have huge specs. It takes me about 20-30 seconds real life time to get +1 grade. So a minute before +2(needed if want extra specs). I estimate my specs to be about 80% higher than normal, at least blood is.
PS- Level 46 amps got the spec treatment too, they are wicked.
I was attracted by the idea and am about to give it a first try ...

Reached G62 by wave 8. Probably because on the map I play one can fully amplify the traps.
Sadly enough the first monsters are beginning to die, despite having like e+35 armor....- apparently not enough.
I will check now and then if somebodyd got the 'nerdpower' to finally reach G100... XD
Upgrading time grows like exponentially, I already realize. So I#m certainly not gonna try to reach the 100.

I'm certainly less experienced than thunder etc. So why that early? I am Lvl 47xx ... More Amplifiers? I upgrade my Gem by duplicating, then adding one pure orange and one specced orange/black ..., amplify by pure black (G64)
That gives me 772.xxx Mana per hit, while tsuhatehk got 427.xxx at G70. Probably just (mainly) the amplifiers...?
If monsters have e35 armor, and based on assumption last endurance wave monsters have e42 hp and e44 regeneration - psorek claims that is the case - than you have only e7 to go :P If I calculated it correctly specials grow 1.7 per gem upgrade, same goes for base dmg. assuming so gem with RBY gem should get 1.7^4 = 8.35 increase in total dmg per hit per grade ( not counting in increase in hits per second or chain length, though I don't think anyone is insane enough to enrage last endurance waves to have more monsters...). That would mean you need another 10 grades and you should be able to beat endurance. Unless my numbers are very wrong grade 80 gem should get you through endurance on the lowest fields.
there is no way monsters with e+35 armor are dying from mana farm traps. You must be mistaken. I currently have the same 5k monsters I've had since wave 5 sitting on a lvl 80 mana trap surrounded by 6 lvl 76 amps. They have e+26 armor.
There is more to optimal amps than you'd expect. Sometimes pure black all around is more powerful, sometimes a combo of a few black/orange and pure black. I'm sure it depends on the hit level. I last experimented with this around lvl 66 in amps and found that having 2 black and 4 B/O gave me the best results.
One thing is for certain, however: amps make a SIGNIFICANT difference. I noticed in this thread a few people setting up mana traps without 6 amps: this is a big mistake. I know there was a lot of talk of how under powered amps are in these forums, but I think that was back when we were using White (which can't be amp'd) instead of black.
If you want to push the limits of this game, find a map that allows room for a trap and 6 amps somewhere near the entrance.
I'm more interested in beating endurance w/o shrines than I am in cores and XP right now, so I don't have any traits set except for orblets, and I'm only enraging the first 5 waves so they're un-killable with the farm
